Metal-responsive elements act as positive modulators of human metallothionein-IIA enhancer activity.

چکیده
The human metallothionein IIA (hMT-IIA) gene contains two enhancer elements whose activity is induced by heavy-metal ions such as Cd2+. To determine the nature of the relationship between the metal-responsive elements and the element(s) responsible for the basal activity of the enhancers, the basal-level enhancer element(s), the hMT-IIA enhancers were subjected to mutational analysis. We show that deletion of the metal-responsive elements had no effect on the basal activity of the enhancer but prevented further induction by Cd2+. On the other hand, replacement of the basal-level enhancer element with linker DNA led to inactivation of the enhancer both before and after treatment with Cd2+. Therefore, the metal-responsive elements seems to act as a positive modulator of enhancer function in the presence of heavy-metal ions. In addition to the two enhancers, the hMT-IIA promoter contained one other element, the GC box, required for its basal expression. Unlike deletion of the basal-level enhancer element, replacement of the GC box with linker DNA had no effect on the ability of the promoter to be induced by Cd2+.

Synergistic induction of promoters containing metal- and glucocorticoid-responsive elements.
We have shown that it is possible to synergistically activate gene transcription when several glucocorticoid responsive elements (GREs) and metal responsive elements (MREs) that coexist within the same promoter are induced simultaneously. To demonstrate this, additional GREs were introduced into a human metallothionein IIA (hMT-IIA) promoter in which some constitutive elements had been deleted....
متن کاملIndividual metal responsive elements of the human metallothionein-IIA gene independently mediate responses to various heavy metal signals.
Metallothioneins (MTs) are small metal-binding proteins that have a role in the defense against heavy metals. Mammalian MT genes are transcriptionally activated by metals such as Cd and Zn through multiple copies of the metal responsive element (MRE) present in the 5'-flanking region. To examine whether each MRE in a single promoter has a distinct role, we characterized seven MREs located upstr...
متن کاملA nuclear factor that recognizes the metal-responsive elements of human metallothionein IIA gene.
Expression of metallothionein (MT) genes is regulated by heavy metals mainly at the transcriptional level, via cis-acting elements called the metal-responsive elements (MREs). A HeLa cell nuclear factor that recognizes MREs of the human MTIIA (hMTIIA) gene, MREBP, was characterized. Mobility shift assay and DNase I footprinting experiments showed that MREBP binds specifically to several MREs pr...

Ursodeoxycholate, used to treat a variety of pathologies, has the ability to reverse cytotoxic and hepatotoxic conditions. We examined HepG2, a hepatic cell line, treated with increasing levels of ursodeoxycholate, for responses of a range of promoters/response elements responsive to DNA damage, heavy metal ions, protein denaturants, aromatic hydrocarbons, retinoids, changes in intracellular AM...

Metallothionein (MT) is a ubiquitous protein with a low molecular weight of 6-7 kDa weight and it was first identified in the kidney cortex of equines as a cadmium (Cd)-binding protein responsible for the natural accumulation of Cd in the tissue. The mammalian MT contains 61 to 68 amino acid residues, in which 18 to 23 cysteine residues are present. The expression of MT starts by binding of met...
